The 145 Downtown / Humber Bay Express bus route operates between the Humber Bay area in south Etobicoke and the Downtown Toronto area, generally in an east-west direction. It also passes within one block of the Queen, Osgoode, St Andrew and King Stations on the Yonge-University-Spadina Subway. Accessible service is provided on the route. Both Queen Station and Osgoode Station are accessible subway stations. Bike racks are available on this route.

Two services are operated. The 145 (Downtown-Kipling) branch operates three eastbound trips in the morning peak period and three westbound trips in the afternoon peak period, from Monday to Friday only. The 145A (Downtown-Royal York) branch operates two eastbound trips in the morning peak period and three westbound trips in the afternoon peak period, from Monday to Friday only.

Eastbound 145 DOWNTOWN EXPRESS / EXTRA FARE
During the morning peak period from Monday to Friday only, these buses operate from Lake Shore Boulevard and Kipling Avenue, or Lake Shore Boulevard and Marine Parade Drive, via east on Lake Shore Boulevard, east on Fleet Street, north on Bathurst Street, east on Adelaide Street, north on Jarvis Street, and west on Richmond Street to Bathurst Street. Eastbound 145 buses operate LOCAL on Lake Shore Boulevard west of the Humber River; stop at Windermere Road, Ellis Avenue, and Colborne Lodge Drive and Bathurst and Fleet; and serve all local stops between Adelaide Street and Peter Street and the downtown area.

Westbound 145 HUMBER BAY EXPRESS / TO KIPLING / EXTRA FARE
During the afternoon peak period from Monday to Friday only, these buses operate from Adelaide Street and Peter Street via east on Adelaide Street, north on Jarvis Street, west on Richmond Street, south on Bathurst Street, west on Fleet Street, south on Fort York Boulevard, west on Lake Shore Boulevard, west and south on Lake Shore Boulevard off-ramp, east on Lake Shore Boulevard, south, west and north on Marine Parade Drive, west on Lake Shore Boulevard to Kipling Avenue. Westbound 145 buses operate LOCAL through downtown, from Adelaide Street and Peter Street to Richmond and John Street; stop at Bathurst Street and Fleet Street, Colborne Lodge Drive, Ellis Avenue, and Windermere Road; and operate LOCAL on Lake Shore Boulevard and Marine Parade Drive west of the Humber River to Kipling Avenue and Lake Shore Boulevard.

Westbound 145A HUMBER BAY EXPRESS / TO ROYAL YORK / EXTRA FARE
During the afternoon peak period from Monday to Friday only, these buses operate from Adelaide Street and Peter Street via east on Adelaide Street, north on Jarvis Street, west on Richmond Street, south on Bathurst Street, west on Fleet Street, south on Fort York Boulevard, west on Lake Shore Boulevard, west and south on Lake Shore Boulevard off-ramp, east on Lake Shore Boulevard, south, west and north on Marine Parade Drive, west on Lake Shore Boulevard, west on Mimico Avenue and south on Royal York Road to Lake Shore Boulevard. Westbound 145 buses operate LOCAL through downtown, from Adelaide Street and Peter Street to Richmond and John Street and; stop at Bathurst Street and Fleet Street, Colborne Lodge Drive, Ellis Avenue, and Windermere Road; and operate LOCAL on Lake Shore Boulevard, Marine Parade Drive, Lake Shore Boulevard, Mimico Avenue and Royal York Road.
